摘要

目的 建立北刘寄奴药材的高效液相色谱法(HPLC)指纹图谱。方法 采用ZORBAX色谱柱(4.6 mm×250 mm,5 μm),乙腈- 0.1%甲酸水溶液梯度洗脱,流速1.0 mL·min-1,检测波长为300 nm,柱温25 ℃,采用LC-MS进行成分分析。结果 建立了北刘寄奴药材的高效液相色谱指纹图谱,标定了21个共有峰,鉴定其中7个共有峰,15 批北刘寄奴药材相似度在0.798到0.992之间,南、北刘寄奴指纹图谱具有显著区别。结论 本法简便,重现性好,可用于区分南、北刘寄奴药材及北刘寄奴药材的质量评价。

Abstract

OBJECTIVE To establish the HPLC fingerprint of Siphonostegia chinensis Benth. METHODS The HPLC fingerprints of 15 batches of Siphonostegia chinensis Benth. were determined on an ZORBAX SB-C18 column (4.6 mm × 250 mm,5 μm) eluted with the mobile phase consisting of 0.1% formic acid solution and acetonitrile in gradient mode at a flow rate of 1.0 mL·min-1; the column temperature was 25 ℃ and the detection wavelength was set at 300 nm.The chemical identification of the fingerprint was done by LC-MS. RESULTS The common mode of the HPLC fingerprint of Siphonostegia chinensis Benth. was set up under the established condition.There were 21 common peaks in the fingerprints of 15 samples, seven of which were identified. The similarities varied from 0.798 to 0.992. There was significant difference in the fingerprints between “Beiliujinu” and “Nanliujinu”. CONCLUSION The method is simple and reliable, which can be readily utilized for distinguishing “Beiliujinu” from “Nanliujinu” and quality control of Siphonostegia chinensis Benth.
